Thermo Scientific™ SuperSignal ELISA Pico Chemiluminescent Substrate is optimized to generate an intense light signal and provide exception performance in luminometer-based assays.

SuperSignal ELISA Pico Substrate provides chemiluminescent detection of horseradish peroxidase (HRP) activity for ELISA methods based on HRP-conjugated antibodies and probes. These ELISAs can take place in either a test tube or a microplate and are quantified by measuring relative light units (RLU) in a luminometer.

We do not recommend diluting the working reagent. It is better to dilute or otherwise optimize primary and or secondary antibody concentrations or use more washes between incubation steps.

The light emission wavelength is read at 425 nm.

When using the SuperSignal ELISA Pico Chemiluminescent Substrate, why are the wells appearing red-orange-yellow in relation to the enzyme?

The substrate solution should remain colorless upon reaction with Horseradish peroxidase (HRP), if the HRP concentration is within the appropriate range. Extremely high concentrations of HRP can cause the substrate color to turn yellow/orange/brown. Alternatively, the color change of the substrate may be due to the presence of some other contaminant.
