25μg supercoiled pZErO-1 (the Zero Background Cloning Kit)or pZErO-2 vector (the Zero Background/Kan Cloning Kit) in TE, 200ng blunt-end test inserts, TOP10 E. coli stab, 25μL T4 DNA ligase, 10X ligation buffer, 1mL sterile water, and 2mL sterile TE. The Zero Background Cloning Kit also contains 1.25mL Zeocin selection agent.

Includes pZErO vector with Zeocin or kanamycin resistance for selection in E. coli

  • pZErO vectors use positive selection to eliminate high background
  • pZErO-1 vector takes advantage of small size of Zeocin resistance gene to reduce the overall vector size for increased ligation and transformation efficiencies
  • pZErO-2 vector includes the kanamycin resistance gene
