Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous APOL1

  • Recognizes human APOL1
  • Human apolipo-protein L1 (APOL1) is a minor component of plasma high density lipoprotein (HDL) particles
    • Thought to be predominantly involved in lipid transport and metabolism
    • Involved in host innate immunity against Trypanosoma parasites
      • Once activated, can lyse parasite and protect human from infection
    • Variants in APOL1 gene associated with chronic kidney disease (FSGS, HIVAN) and hypertensive nephropathy

Process:

  • Antibody specific for APOL1 pre-coated onto microwells
  • APOL1 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for APOL 1 is added to detect captured APOL1 protein
  • For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
