Description
- Recognizes human APOL1
- Human apolipo-protein L1 (APOL1) is a minor component of plasma high density lipoprotein (HDL) particles
- Thought to be predominantly involved in lipid transport and metabolism
- Involved in host innate immunity against Trypanosoma parasites
- Once activated, can lyse parasite and protect human from infection
- Variants in APOL1 gene associated with chronic kidney disease (FSGS, HIVAN) and hypertensive nephropathy
Process:
- Antibody specific for APOL1 pre-coated onto microwells
- APOL1 protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody specific for APOL 1 is added to detect captured APOL1 protein
- For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
