The pYSG-IBA vector allows the high-level expression of proteins in yeast.
- The vector carries the copper inducible promoter (CUP1) for controlled high-level expression.
- The expressed recombinant protein will be localized in the cytoplasm.
- High-level expression in yeast with high copy number
- Favors correct protein folding and the production of soluble proteins - inclusion bodies rarely form
- Tightly regulated expression due to Cu++-inducible CUP1 promoter
- Ampicillin resistance gene and markers Leu2-d and URA3 in every vector
- Leu2-d auxotrophy marker for selection to increase plasmid copy number during expression
- URA3 auxotrophy marker for selection after transformation
Please note that cloning into pYSG-IBA Acceptor Vectors compulsorily requires the restriction enzyme Esp3I since no other MCS for the integration of a gene of interest is available. In addition to the direct cloning of the gene of interest into pYSG-IBA vectors with Esp3I, another option via a so-called Entry-Vector is possible.
