Description
When packing a column for use in low-pressure flash chromatography, users must ensure that the adsorbent cannot be flushed from the column and that the adsorbent does not become suspended in the eluent. Sea sand is placed above and below the silica to ensure a stable gel bed.
- A pad of glass fiber wadding is placed in front of the tap and a layer of sea sand (about 0.5–1.0cm) is filled in above it
- Then the dry adsorbent is packed into the column and is settled down until it forms a stable gel bed
- The silica bed is covered with another layer of sea sand (about 0.5–1.0cm) to avoid suspension of the silica surface
