Mouse Monoclonal Antibody

Description: The monoclonal antibody SAM11 recognizes human PAR2 (Protease activated receptor 2). PAR2 is a member of the PAR family which is a sub-family of the G protein coupled receptors (GPCR). Unlike most of the GPCR family members, PARs are not activated through ligand binding, but through proteolytic cleavage of the amino terminus. PAR2 activation is induced by trypsin, mast cell tryptase, neutrophil proteinase 3, tissue factor-factor VIIa-factor Xa, human kallikrein-related peptidases and membrane-tethered serine proteinase-1/matriptase-1. Unlike other PARs, PAR2 is not activated by thrombin. PAR2 is widely expressed in many tissues, including skin, liver, kidney, small intestine, and colon. PAR2 plays a key role in numerous inflammatory diseases and cancers. The SAM11 antibody has been reported to inhibit the ability of serine proteinases to activate PAR2. Applications Reported: This SAM11 antibody has been reported for use in immunohistochemical staining of formalin-fixed paraffin embedded tissue sections, microscopy, ELISA, functional assays, and immunocytochemistry. Applications Tested: This SAM11 antibody has been tested by immunohistochemistry of formalin-fixed paraffin embedded human tissue using low or high pH antigen retrieval. This SAM11 antibody has also been tested by immunocytochemistry of both methanol and formaldehyde-fixed and permeabilized cells and can be used at less than or equal to 10 μg/mL.

Coagulation factor II (thrombin) receptor-like 1 (F2RL1) is a member of the large family of 7-transmembrane-region receptors that couple to guanosine-nucleotide-binding proteins. F2RL1 is also a member of the protease-activated receptor family. It is activated by trypsin, but not by thrombin. It is activated by proteolytic cleavage of its extracellular amino terminus. The new amino terminus functions as a tethered ligand and activates the receptor. The F2RL1 gene contains two exons and is widely expressed in human tissues. The predicted protein sequence is 83% identical to the mouse receptor sequence.
