  • 1X OPTIZYME Buffer 2: 10mM Tris-HCl (pH 7.5 at 37°C), 10mM MgCl2, 50mM NaCl and 0.1mg/ml BSA
  • Incubate at 55°C
Enzyme Activity in OPTIZYME buffers:
  • Buffer 1: 20 - 50%
  • Buffer 2: 100%
  • Buffer 3: 20 - 50%
  • Buffer 4: 100%™
  • Buffer 5: 20 - 50%
™Star activity appears at a greater than 5-fold over-digestion
Storage Buffer:
  • 10mM Tris-HCl (pH 7.4 at 25°C), 100mM KCl, 1mM DTT, 1mM EDTA, 0.2mg/ml BSA and 50% (v/v) glycerol
  • Ligation and Re-cleavage:
  • More than 95% of the DNA fragments can be ligated and re-cut after 10-fold over-digestion with BclI.
  • Notes:
    • Incubation at 37°C results in 50% activity.
    • Greater than 15-fold overdigestion with BclI may result in star activity.
    • Assayed using lambda DNA (dam-)
    • BclI is blocked by dam methylation. To avoid dam methylation, use a dam-, dcm- strain.
