Description: This BuGR2 monoclonal antibody reacts with mouse glucocorticoid receptor NR3CR1. This clone has been also reported to cross-react with human NR3CR1. This BuGR2 antibody is recommended to be used with the Foxp3/Transcription factor staining buffer set (Product No. 00-5523-00). Applications Reported: This BuGR2 antibody has been reported for use in flow cytometric analysis. This clone has been also reported to cross-react with Human, Non-human primate, Rabbit, Rat, Sheep, Xenopus, Yeast NR3CR1. Applications Tested: This BuGR2 antibody has been tested by flow cytometric analysis of stimulated mouse splenocytes using the Foxp3/Transcription Factor Staining Buffer Set (Product No. 00-5523-00) and protocol. Please refer to Staining Intracellular Antigens for Flow Cytometry, Protocol B: One step protocol for intracellular (nuclear) proteins located at www.thermofisher.com/flowprotocols. This may be used at less than or equal to 0.125 μg per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. Light sensitivity: This tandem dye is sensitive to photo-induced oxidation.

Please protect this vial and stained samples from light. Fixation: Samples can be stored in IC Fixation Buffer (Product No. 00-8222-49) (100 μL of cell sample + 100 μL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (Product No. 00-5333-57) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically. Excitation: 488-561 nm; Emission: 775 nm; Laser: Blue Laser, Green Laser, Yellow-Green Laser Glucocorticoid receptor (GR) is encoded by the gene NR3C1, and can function both as a transcription factor and as a regulator of other transcription factors. This receptor is typically found in the cytoplasm, but is transported into the nucleus upon ligand binding. GR is involved in inflammatory responses, cellular proliferation, and cell differentiation. Mutations in this gene are associated with generalized glucocorticoid resistance. T-cells are a critical cellular target of GR, as immune activation in mice lacking GR resulted in significant mortality. This lethal activation is rescued by PTGS2 (COX-2) inhibition but not steroid administration or cytokine neutralization. All normal mammalian tissues examined to date have been shown to contain glucocorticoid receptor.
